The CD4/CD8 Lymphocyte Enumeration Test is a specialized flow cytometry blood test that measures the number of CD4+ (T-helper) and CD8+ (cytotoxic T) lymphocytes in the blood. These are key subtypes of T-cells, which are white blood cells essential to the immune system’s ability to fight infections and monitor abnormal cells.
This test also calculates the CD4/CD8 ratio, which provides insight into immune system balance and function. It is a critical tool for evaluating immune status in various medical conditions, especially HIV/AIDS.

Description

Why It’s Done:

This test is performed to:
• Monitor HIV infection and determine disease progression by assessing the immune system’s status.
• Evaluate response to antiretroviral therapy (ART) in HIV-positive patients.
• Detect immune suppression or dysfunction in other conditions such as:
o Primary immunodeficiencies
o Autoimmune diseases
o Certain cancers (e.g., lymphomas)
o After organ or bone marrow transplantation
• Investigate chronic infections or recurrent illness where immune dysfunction is suspected.
A low CD4 count, high CD8 count, or abnormal CD4/CD8 ratio can indicate a compromised immune system.

Preparation:

• No fasting or special dietary restrictions are required.
• Inform your healthcare provider of:
o Any medications being taken, especially immunosuppressants, steroids, or chemotherapy drugs.
o Recent infections, vaccinations, or medical procedures, which may affect results.
• A blood sample is collected from a vein in your arm for analysis via flow cytometry.
