The ANA (Anti-Nuclear Antibody) – IFA (Indirect Immunofluorescence Assay) test is a blood test that detects autoantibodies that target substances found in the nuclei of cells. It is considered the gold standard for ANA testing due to its high sensitivity.
In this test, patient serum is applied to cells fixed on a slide, and if ANA are present, they bind to nuclear components. A fluorescent dye is then added, which attaches to these antibodies and glows under a fluorescence microscope. Different staining patterns can provide clues to specific autoimmune diseases.

    Why It’s Done

    The ANA – IFA test is mainly used to:
    • Help diagnose autoimmune diseases, especially when symptoms suggest:
    o Systemic Lupus Erythematosus (SLE)
    o Sjögren’s Syndrome
    o Scleroderma
    o Mixed Connective Tissue Disease
    o Polymyositis/Dermatomyositis
    • Evaluate unexplained symptoms such as:
    o Persistent joint pain or swelling
    o Chronic fatigue
    o Skin rashes (especially butterfly-shaped rash on the face)
    o Recurrent fevers
    • Monitor disease activity or response to treatment in diagnosed autoimmune conditions

    Preparation for the Test

    • Fasting: Not required.
    • Medications: Inform your healthcare provider about any medications or supplements, as some can influence immune system activity.
    • No special preparation is generally needed.
