The Albert Stain test is a microbiological staining technique used to identify Corynebacterium diphtheriae, the bacterium responsible for diphtheria. This test involves staining a sample (usually from the throat or a nasal swab) with Albert stain, which highlights specific structures within the bacteria—particularly metachromatic granules—that help differentiate C. diphtheriae from other bacteria.

Description

Why It’s Done:

The Albert Stain test is performed to:
• Detect Corynebacterium diphtheriae in suspected cases of diphtheria
• Confirm diagnosis in patients with symptoms like:
o Sore throat
o Thick gray or white coating in the throat
o Difficulty breathing or swallowing
o Swollen neck glands (“bull neck”)
• Aid in public health surveillance during outbreaks of diphtheria
It is a quick and cost-effective test that provides preliminary identification before confirmation by culture or PCR.

Preparation:

• No special preparation is required for the patient.
• A throat swab or nasal swab is usually collected by a healthcare professional using sterile technique.
• Patients should avoid eating or drinking 15–30 minutes prior to sample collection for optimal results.
